چکیده

JCB • VOLUME 189 • NUMBER 6 • 2010 922 J oan Brugge was an undergraduate at Northwestern University when her sister was diagnosed with a brain tumor. Her sister’s doctors couldn’t say what caused the disease; they speculated that a virus might be involved. This spurred Brugge’s lifelong quest to learn about the causes of cancer, starting with the study of tumor viruses (1). Brugge identifi ed the protein encoded by the src transforming gene of Rous sarcoma virus and its cellular homologue as a postdoc in Ray Erikson’s lab at the University of Colorado (2). She further characterized the cellular Src protein during her time as a group leader at Stonybrook and the University of Pennsylvania (3). Later, following a brief stint in the private sector, she set up shop at Harvard Medical School to study the cellular processes that drive the striking morphological and phenotypic changes that accompany cancerous transformation in humans (4–6). A leader in the fi eld of cancer biology, Brugge manages a vibrant research program while meeting the demands of chairing the Department of Cell Biology. But, she took time out to chat with us about her career—past, present, and future.

Q&A: Joan Brugge on empowering post-docs.

When we were studying the YAP oncogene, we noted that down-regulation of YAP caused an exceptionally dramatic phenotype during cytokinesis. That was diffi cult to ignore, so we initiated a project to understand how YAP controls cytokinesis to provide insight into its function. In another case, we observed that when cells were detached from their extracellular matrix, one cell would literally in...

At the leading edge of cancer research: an interview with Joan Brugge

college in the 1960s, scientists could only speculate about what caused cancer. Evidence was accumulating to suggest that viruses caused the disease, but the lack of molecular techniques at the time limited scientists from probing the underlying pathological mechanisms. A penchant for problem solving and a fascination with experimentation led Joan Brugge to pursue research questions in cancer d...

Uncovering a Tumor Suppressor for Triple-Negative Breast Cancers

"Triple-negative" breast cancers are aggressive malignancies that respond poorly to treatments. Now Sun et al. (2011) find that the activity of the protein tyrosine phosphatase PTPN12 is lost in a large percentage of this breast cancer subtype, offering molecular drivers and possible therapeutic targets for this heterogeneous and intractable cancer.

Altered metabolic requirements in cancer cell migration and metastasis

Background Metastasis poses a great challenge in clinical management of many cancers. Metabolic demands of cancer cell proliferation – i.e. elevated aerobic glycolysis for biomass generation – have been well characterized, but the contribution of altered metabolism to metastasis remains to be elucidated. While elevated aerobic glycolysis, a phenomenon termed Warburg effect, is a hallmark of pro...

IL-6 involvement in epithelial cancers.

In this issue of the JCI, two reports provide intriguing new information on the role of the inflammatory cytokine IL-6 in breast and lung cancer. The study by Sansone et al. implicates IL-6 in the instigation of malignant properties in breast cancer stem cells (see the related article beginning on page 3988). The study by Gao et al. identifies mutant variants of EGFR as inducers of IL-6 in lung...
